The effect of miR-122-5p on the inter-Sertoli TJ permeability barrier in vitro. The miR-122-5p mimic and inhibitor were transfected into SCs. The functionality of SCs on the TJ barrier was quantified as the TER across the Sertoli cell epithelium on Matrigel-coated bicameral units, and the TER was measured on days 1, 2, 3, 4, 5, 6, 7 and 8. There were no differences between the three groups from day 1 to day 3 (p > 0.05). No difference was found between the blank group and miR-122-5p inhibitor group from day 4 to day 8 (p > 0.05). However, the TER of the miR-122-5p mimic group was significantly lower than the other two groups beginning on day 4 (p < 0.05). This result suggests that the miR-122-5p mimic inhibits the assembly of the inter-Sertoli TJ permeability barrier in vitro.
